For using Snapdragon profiler, Should Mobile and PC are connected same Wi-Fi AP?

Could I run the program when only connected USB cable?

If you connect via a USB cable (which is recommneded) it shouldn't matter whether your smartphone and comptuer are connected to the same wireless AP. If you connect via USB over Wi-Fi, you'll obviously need to be connected to the same network.
